摘要
目的:研究清脑宣窍滴丸对急性脑缺血/灌注损伤引起的一氧化氮(NO)含量和一氧化氮合酶(NOS)活力的改变及机制。方法:采用线栓法致大鼠大脑中动脉缺血再灌注(MCAO)模型,测定大鼠手术侧脑匀浆中NO含量及结构型一氧化氮合酶(cNOS)、诱导型一氧化氮合酶(iNOS)、总NOS活力。结果:清脑宣窍滴丸45,90,180 mg.kg-1可明显降低MCAO大鼠患侧脑匀浆中NO含量,180 mg.kg-1可明显降低术侧大脑组织升高的总NOS,cNOS和iNOS活力(P<0.05);90 mg.kg-1也可明显降低升高的总NOS活力(P<0.01)。45 mg.kg-1明显降低升高的总NOS活力(P<0.05)。结论:清脑宣窍滴丸可能通过减少NO含量,降低iNOS的活性来防治脑缺血/再灌注损伤。
Objective: To study the role and mechanism of Qing-nao-xuan-qiao Dropping pill(QNXQD) on acute cerebral ischemie injury. Methods: The middle cerebral artery occlusion (MCAO)model induced by inserting a monofilament suture was adopted. The content of NO, and the activity of eNOS, iNOS, total NOS in brain homogenate was determined. Results: QNXQD 45,90,180 mg· kg^-1, can significantly reduce the content of NO in the rat brain homogenate of the troubled side; 180mg· kg^-1 can significantly decrease the increased eNOS, iNOS; total NOS activity in the side of the brain surgery ( P 〈 0.05) ; 90 mg· kg^-1 can significantly decrease the increased total NOS activity ( P 〈 0.01 ) ; and 45mg· kg^-1 can significantly decrease the increased total NOS activity( P 〈 0.01 ). Conclusion: QNXQD has protective effect on cerebral isehemia and the possible mechanism may be related to reducing content of NO, decreasing the activity of cNOS, iNOS, total NOS.
