膝骨关节炎患者尿液代谢谱与中医证候的相关性研究

Correlativity Between Urinary Metabolite Profiles and Chinese Medical Syndromes in Knee Osteoarthritis Patients

目的:比较膝骨关节炎(OA)患者与正常对照者之间尿液代谢谱的差异,及膝OA不同证型间代谢表型的差异,为中医"证"的本质研究提供客观依据。方法:利用气相色谱-质谱(GC-MS)联用技术检测37例膝OA患者和37例正常对照者尿液代谢产物,识别OA潜在的生物标记物;比较分析OA两个证型组的尿液代谢谱变化。结果:OA组与正常对照组尿液中代谢物的总离子流图存在明显差异,两证型组尿样中多种内源性代谢物的含量和尿液代谢谱差异明显。结论:中医"证"的本质具有特定的生化物质基础;代谢组学分析可以为OA的临床诊断和中医"证"本质研究提供新的技术手段。

Objective： To compare the differences of endogenous metabolites in urine from the patients suffering from knee osteoarthritis（KOA） and the healthy people, and between two Chinese syndromes of OA of metabolic phenotypes. Methods： Combined gas chromatography and mass spectrometry（GC-MS） method was utilized to detect the metabolic products in the urine sampies of 37 KOA patients and 37 healthy people, identify some potential biomarkers and metabolite profile of the two syndromes. Results ： The contents of many kinds of endogenous metabolites in the urine samples of KOA patients and healthy people were significantly different ; the contents of endogenous metabolites and metabolic profiles were of great differences between two syndromes. Conclusion ： Chinese medical syndrome has its biochemical substances and the technique of metabonomics can be used for OA diagnosis and determining the essence of Chinese medical syndromes.
