复黄生肌愈创油膏对糖尿病模型创面TGF-β_1与smad3、smad7水平的动态影响 被引量:4

摘要 目的:探讨复黄生肌愈创油膏(简称复黄膏)促进创面愈合的作用机制。方法:36只雄性Wistar大鼠随机分为正常对照组、糖尿病模型组、复黄膏治疗组。复制Wister大鼠糖尿病创面模型,药物干预后分第3天和第11天两个观察时段,采用免疫组化技术观察不同时段创面中TGF-β_1、smad3、smad7含量变化。结果:第3天时,复黄膏治疗组TGF-β_1含量、smad3含量均明显高于模型组(P<0.05),但与正常组比较无统计学差异;smad7含量3组间比较无统计学差异。第11天时,复黄膏治疗组TGF-β_1含量、smad3含量均明显低于正常组(P<0.05),但与模型组比较无统计学差异;复黄膏治疗组smad7含量明显高于正常组和模型组(P<0.05)。结论:TGF-β_1、smad3在复黄膏治疗组创面愈合早期表达上调,表明复黄生肌愈创油膏通过促进创面生长因子的表达及其信号转导,从而起到促进创面愈合的作用;而在创面愈合晚期, TGF-β_1、smad3在复黄膏治疗组表达下调,smad7作为细胞内Smad信号的抑制剂表达上调,可能为复黄生肌愈创油膏促进了创面愈合的负反馈机制,及时中止成纤维细胞过度生长,防止瘢痕形成。 Objective: To investigate the mechanism of " Fuhuang Shengji Yuchuang Ointment" (FSYO) in promoting the wound healing. Methods : Thirty-six male Wistar rats were randomized into 3 groups : normal control group, diabetic model group and FSYO group. The model of wound in granulation tissue was established, and the expression of TGF-β1, smad3 and smad7 were detected with immunohistochemistry on the third day and eleventh day after the drug intervention. Results: On the third day, the expression of TGF-β1 and smad3 in FSYO group were higher than that of model group ( P 〈0.05 ), but when compared with the normal control group there was no statistical difference. There was no statistical difference in the expression of smad7 among all the three groups. On the eleventh day, the expression of TGF-β1 and smad3 in FSYO group were lower than those of normal control group ( P 〈0.05 ), when compared with the normal control group it was of no statistical difference. The expression of smad7 in FSYO group was higher than those of the other two groups (P 〈0.05). Conclusion: The expression of TGF-β1 and smad3 in FSYO group were up-regulated at the early stage of wound healing, which indicates that the FSYO can promote wound healing via spurring the expression of certain cytokines and their signal transduction ; at the late stage of wound healing, the expression of TGF-β1 and smad3 in FSYO group are down-regulated, as well as the expression of smad7, the inhibitor of smads in cell signal transduction, are up-regulated, which reflects that FSYO may open the inverse signal feedback of wound healing, thus to prevent the scars formation by stopping the excessive cells proliferation.
