摘要
目的建立北豆根有效部位的反相高效液相色谱鉴别和有效成分含量分析方法。方法选用Agilent C_(18)(250 mm×4.6 mm,5μm)柱,乙腈-0.05%三乙胺水溶液为流动相,梯度洗脱,流速为1.0 ml/min,检测波长为282 nm。结果13批不同来源的北豆根有效部位的HPLC指纹图谱中可检出12个相对位置稳定的色谱峰,可以此作为鉴别的指标峰,蝙蝠葛苏林碱、蝙蝠葛碱的含量分别为2.19~16.35、2.32~10.56 mg/g。结论本实验分析方法具有较强的针对性和准确性,可用于北豆根及其制剂的质量控制。
Objective To establish a qualitative and quantitative method with HPLC for controlling the quality of Menispermum dauricum DC.. Methods The experimental conditions of the HPLC method were as follows: Agilent Cls column (250 mm×4.6 mm, 5μm), mobile phase of C H3 CN-0.05 % C6 H15 N, flow rate at 1.0 ml·min^-1 . detection wavelength at 282 nm. Results Under the qualitative conditions, the 12 common peaks in HPLC fingerPrint of the active part from 13 different samples could be used as index peaks for qualitative identification. In HPLC quantitative analysis, the contents of Daurisoline and Dauricine in Menispermum dauricum DC. were 2.19 - 16.35 mg·g^-1 and 2.32-10.56 mg·g^-1 respectively. Conclusion The analytical method with qualitative fingerPrint of active part and quantitative measurement of active components can accurately be used for controlling the quality of Menispermum dauricum DC. and its preparations.
