摘要
目的:评估热籽对荷瘤大鼠肿瘤加温治疗的杀伤效果。方法:将接种Walker-256肿瘤细胞的Wistar大鼠随机分为5组,C组(空白对照组)、M组(磁场对照组)、T组(热籽对照组)、H组(加温治疗,H1组:瘤内植入2颗居里点57℃热籽,持续30 min;H2组:2颗居里点70℃热籽,持续6 min)。在加温治疗后即刻和24 h取H组及C组肿瘤组织行组织学及免疫组织化学检查,治疗后第9天测5组肿瘤体积及质量。结果:加热后热籽周围肿瘤组织呈现凝固性坏死,热休克蛋白70表达较C组增加,24 h后表达明显减少。H1组、H2组热疗后肿瘤体积与C组、M组比较差异显著(P<0.05),肿瘤质量与C组差异显著(P<0.05)。结论:热籽加温治疗对荷瘤大鼠肿瘤生长有一定的抑制作用。
Objective: To evaluate the killing effect of thermoseed induced heating on implanted tumor in rats. Methods: Wistar rats with tumor Walker-256 were divided into five groups, including group C (blank control group), group M (magnetic control group), group T (thermoseeds control group) and heating treatment groups: group H1 and group H2. In group H1, two thermoseeds with 57 ℃ curie point were implanted into tumor tissues of each rat with a heating time of 30 minutes. For group H2 , two thermoseeds with 70℃ curie point were implanted into tumor tissues of each rat with a heating time of 6 minutes. Histological and immunohistoehemistry observations on tumors were carried out in groups H1, H2 and C immediately and in 24 hours after the heating treatment. Tumor volumes and weights were measured in all the five groups in 9 days after the treatment. Results: Tumor tissues around thermoseed array showed coagulation necrosis after heating. Heat shock protein 70 expression was stronger in group H1 and group H2 than in group C, while the expression decreased in 24 hours later. After the treatment, the tumor volumes in groups H1 and H2 were significantly smaller than those in groups C and M (P 〈0.05 ) , and tumors in groups H1 and H2 were significantly lighter than those in group C (P 〈 0.05 ). Conclusions: Thermoseeds induced heating could inhibit tumor growth in rats.
