血晶素诱导血红素氧合酶-1在脑外伤中的表达及意义

Role of hemin-induced heme oxygenase-1 expression in rat central nervous system after brain trauma

摘要 目的探讨血红素氧合酶-1(HO-1)在颅脑外伤中的作用。方法48只SD大鼠按照随机数字表法分为血晶素处理组、生理盐水组和假手术组。用液压冲击伤复制颅脑外伤的大模型.伤后予以45mg/100mg体质量血晶素、生理盐水腹腔注射干预H0—1的表达。假手术组只切开头皮,颅骨钻孔,其他不作任何处理。斜坡试验观测大的行为学改变,尾静脉注射伊文思兰测定血脑屏障的通透性,SP免疫组织化学测定H0-1的表达。结果血晶素处理组HO—1的表达[(125.52±14.39)个]较生理盐水组[(100.63±12.32)个]和假手术组[(79.37±12.89)个]明显增加,差异有统计学意义(P〈0.05)。血晶素处理组大鼠斜坡试验时间[(7.38±1.69)s]较生理盐水组[(10.01±1.61)s]缩短,差异有统计学意义(P〈0.05)。血晶素处理组血脑屏障的通透性较生理盐水组明显降低.差异有统计学意义(P〈0.05)。结论HO-1在颅脑外伤中的表达对中枢神经系统具有保护作用。 Objective To explore the role of heme oxygenase-1 (HO-1) in the central nervous system after brain trauma. Methods Forty-eight adult SD rats were randomized into brain trauma+heroin group, brain trauma+normal saline group and sham-operated group, and in the former two groups, the rats were subjected to fluid percussion injury of the brain followed by intraperitoneal injections of 45 mg/100 mg heroin or saline accordingly. The rats in the sham-operated group received incision of the scalp and drilling on the skull only. The behavioral changes of the rats after the treatments were evaluated by testing the latency of head turning on a ramp. The permeability of the blood brain barrier (BBB) was assessed by measuring the brain tissue content of Evans blue injected into the tail vein, and the expression of HO-1 in the brain tissue was detected using immunohistochemistry with SP method. Results HO-1 expression in the brain tissue of hemin-treated rats (125.52±14.39) was significantly increased as compared with that of normal saline-treated rats (100.63±12.32) and sham-operated rats (79.37±12.89) (P〈0.05). The latency of head turning on the ramp was significantly shorter in hemin-treated group (7.38±1.69 s) than in normal saline group (10.01±1.61s, P〈0.05). The permeability of the BBB was markedly decreased in heroin group. Conclusion HO-1 expression in the brain tissue may provide protection of central nervous system after brain trauma.
