摘要
目的探讨2型糖尿病(T2DM)患者脂肪酸结合蛋白(FABP2)及载脂蛋白E(ApoE)与冠心病(CHD)的关系。方法采用聚合酶链反应-限制性片段长度多态性(PCR-PFLP)技术分别检测T2DM并发CHD组及T2DM非CHD组FABP2、ApoE基因型,并检测T2DM并发CHD组不同等位基因携带者血清甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HLD-C)、低密度脂蛋白胆固醇(LDL-C)。结果T2DM并发CHD组FABP2A/T突变频率高于非冠心病组(P<0.05),T等位基因携带者与A等位基因携带者相比TG、TC、LDL-C浓度升高,血HDL-C浓度降低,有显著性差异(P<0.05);T2DM并CHD组ε4等位基因频率明显高于对照组(P<0.01),ε4等位基因携带者与ε3等位基因携带者相比TC、LDL-C浓度升高,HDL-C浓度降低,差异有统计学意义(P<0.05),FABP与ApoE基因连锁分析,FABP2T基因和ApoEε4基因连锁时,T2DM并发CHD的几率增加(P<0.05)。结论FABP2T基因及ApoE/ε4等位基因增高T2DM并发CHD的遗传易感性。
Objective To study the relationship of fatty acid binding protein-2 (FABP2) and apolipoprotein E (ApoE) with coronary heart disease (CHD) in type-2 diabetes mellitus (T2DM) patients. Methods FABP2 and ApoE gene polymorphisms were detected by PCR-RFLP. At the same time all CHD in T2DM patients' serum triglyceride (TG), total cholesterol (TC), high-density lipoprotein cholesterol were (HDLC), low-density lipoprotein cholesterol (LDLC) were detected. Results The T-allele frequencies of FABP2 were higher (P〈0.05), the levels of serum TC, TG and LDLC of T-allele carriers were higher while HDLC was lower than that of A-allele carriers (P〈0.05). The ε3 -allele frequencies of ApoE were higher (P〈0. 01), the levels of serum TC and LDLC of T-allele carriers were higher, but HDLC was lower than that of ε3-allele carriers (P〈0.05). Combined analysis of FABP2 and APOE genotype showed that the FABP2T +/APOEε4+ genotype was a risk factor of CHD in T2DM patients (P〈0.05). Conclusion FABP2T and ApoEε4 alleles increse the susceptibility to CHD in T2DM.
