摘要
目的研究肝癌组织中Survivin和PTEN基因表达,探讨两种基因和其编码的蛋白产物之间的相互关系以及它们与肝癌临床病理因素的关系。方法RT-PCR法检测28例肝癌组织和其相对应的癌旁组织中Survivin和PTEN基因mRNA的表达情况,免疫组化方法检测51例肝癌组织石蜡切片中Survivin和PTEN蛋白的表达。分析两者mRNA和蛋白的表达情况与肝癌临床病理因素的关系。结果28例肝癌组织中Survivin mRNA表达率为46.43%(13/28),高于癌旁组织0%(0/28)(P<0.05);PTEN mRNA在癌组织中表达率分别为35.71%(10/28),低于癌旁组织71.43%(20/28)(P<0.05)。Survivin mRNA的表达与肝癌病理分级和癌栓形成有关,PTEN mRNA的表达与肝癌病理分级有关(P<0.05)。Survivin蛋白在肝癌组织中阳性表达率为64.70%(33/51),与肝癌的病理分级有关。PTEN蛋白在肝癌组织中阳性表达率为47.06%(24/51),与肝癌的病理分级和癌栓形成有关。结论高表达Survivin的肝癌细胞常分化差,易转移。PTEN在肝癌细胞中表达显著减少或缺失,与癌的分化和转移密切相关。肝癌时Survivin激活与上调,PTEN失活、下调或缺失,共同参与调控肝癌分化和转移。
Objective To observe the expressions of PTEN and Survivin gene in HCC, investigating relationship between the two genes mRNA and their protein expressions, and their relationship with clinicopathological factors. Methods Survivin and PTEN gene mRNA ex- pressions were detected in 28 cases with HCC and their adjacent noncancerous tissues by RT-PCR, their protein was detected in 51 cases with HCC by immunohistochemistry (IHC). To analyze the relationship between the two genes mRNA and protein expressions and their relationship with clinicopathological factors. Results RT-PCR results showed that positive rate of Survivin gene mRNA expressions in HCC was 46.43% (13/28) , higher than that in adjacent tissues 0% (0/28)(P 〈0.05). Positive rate of PTEN gene mRNA expressions in HCC was 35.71% ( 10/28 ) , lower than that in adjacent tissues 71.43% (20/28) ( P 〈 0. 05). Survivin gene mRNA expressions were associated with pathological grading and vein micro-metastasis in HCC. PTEN gene mRNA expressions were associated with pathological grading in HCC- IHC results showed positive rate of Survivin protein expressions in HCC were 64.71% (33/51), which were associated with pathological grading in HCC. Positive rate of PTEN protein expressions in HCC were 47.06% (24/51) , which were associated with pathological grading and vein micro-metastasis in HCC. Conclusion Survivin is closely associated with differentiation and metastasis in HCC. PTEN expression decreased or vanished in HCC compared with adjacent noncancerous tissues, and closely associated with differentiation and metastasis in HCC. In HCC developing, there was Survivin activating and up-regulating with PTEN inactivating and down-regulating. The two genes regulated differentiation and metastasis of HCC.
