非小细胞肺癌组织HIF-1α和CAIX及VEGF表达临床意义的探讨 被引量:11

Expressions of HIF-1α,carbonic anhydrase Ⅸ and VEGF in non-small cell lung carcinoma and their clincical significance

摘要 目的:探讨非小细胞肺癌(non-small cell lung cancer,NSCLC)病理组织中乏氧诱导因子-1α(hypoxia inducible factor 1α,HIF-1α)、碳酸酐酶9(carbonic anhydraseⅨ,CAⅨ)和血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达水平及其临床意义。方法:免疫组化方法测定59例NSCLC和14例肺部良性病变病理切片中CAⅨ、HIF-1α和VEGF的表达,并与患者的临床资料进行统计分析。结果:59例肺癌组织中27例(45.8%)有HIF-1α表达,44例(74.6%)有CAⅨ表达,43例(72.9%)有VEGF表达。CAⅨ、HIF-1α和VEGF的表达在不同临床分期、不同瘤体大小差异均无统计学意义;CAⅨ的表达在不同病理类型差异有统计学意义,鳞癌中CAⅨ的表达显著高于腺癌,P=0.039;HIF-1α的表达与组织分化程度呈负相关,P=0.015;VEGF的表达在淋巴结转移阳性组较阴性组高,P=0.031。高HIF-1α表达与高CAⅨ表达有显著相关性,P=0.004,其表达区域重叠。同时,HIF-1α和VEGF的阳性表达相关,P=0.011。结论:HIF-1α和CAⅨ均在NSCLC中高水平表达,均有可能影响NSCLC的生物学行为和预后。 OBJECTIVE: To investigate the correlation between the expressions of hypoxia inducible factor 1α (HIF-1α), carbonic anhydrase Ⅸ(CA Ⅸ) and vascular endothelial growth factor (VEGF) in human non small cell lung cancer and their clinical significance. METHODS: Immunohistochemistry was used to detect the expressions of CAⅨ, HIF-1α and VEGF in pathological tissue sections and the correlation of them with the age, sex, size of tumor, histopatho logical type,differentiation,stage and lymph node metastasis were an alyzed statistically. RESULLTS: HIF-1α was expressed in 27 eases (45.8%),CAIX in 44 cases (74.6%), and VEGF in 43 cases (72. 9%). There was no significant difference among stages and tumor sizes. The expression of CAⅨ among the patients with differ ent histopathological types were different significantly, P = 0. 039. The expression of HIF-1α among the patients with different differen ciations were different significantly, P=0. 015. The expressions of VEGF between the patients with or without lymoh node metastasis were significantly different, P = 0.031. HIF-1α expression was strongly correlated with CAⅨ and co-localized with CAⅨ in corre sponding areas, P=0. 004. HIF-1α expression was also significantly correlated with VEGF, P=0. 011. CONCLUSION:The high expressions of CA Ⅸ and HIF-1α occurre in most of cases of NSCLC, which may play a role in the biologic behavior and prognosis of NSCLC.
