子宫内膜、正常早孕和自然流产蜕膜组织中△Np63的表达

摘要 目的观察△Np63蛋白在自然流产蜕膜组织中的表达,探讨△Np63在胚泡植入和早期妊娠维持中的作用。方法收集自然流产的蜕膜27例组织(其中胚胎停止发育23例、习惯性流产1例及不全流产3例),正常早孕蜕膜66例,非妊娠期子宫内膜组织57例(其中增生期33例,分泌期24例),用SP免疫组化染色法观察△Np63的表达。结果△Np63广泛表达于腺上皮和间质细胞中。增生期和分泌期子宫内膜中△Np63主要表达在腺上皮,蜕膜组织中△Np63在间质蜕膜细胞中的有较强的表达。流产组△Np63蛋白阳性率为55.6%,明显低于增生期的90.9%、分泌期的87.5%及正常早孕组的86.4%(P<0.05)。△Np63高表达率在分泌期组及流产组分别为16.7%和11.2%,明显低于增生期的63.6%和正常早孕组的57.6%(P<0.01)。结论△Np63可能参与子宫内膜的生理周期变化,并与子宫内膜向早孕蜕膜组织转化及维持早孕关系密切。 Purpose To observe △Np63 protein expression in decidua of spontaneous abortion and to discuss its function in the implantation of blastocyst and the maintenance of early pregnancy. Methods Twenty-seven cases of decidua of spontaneous abortion ineluding twenty-three cases of embryos ceasing to develop, one case of recurrent abortion and three cases of incomplete abortion, sixtysix cases of deeidua of normal early pregnancy and fifty-seven cases of human endometria in non-pregnant period ( 33 at proliferative phase and 24 at secretory phase) were collected for this study. The expression of △Np63 was detected by SP immunohistoehemistry. Results △Np63 protein was extensively distributed in glandular epithelia and stromal cells. During proliferative and secretory stages,△Np63 protein was mainly located in glandular epithelia, while its increased expression was found in decidua stromal cells during early pregnancy. The positive rate of △Np63 in group of spontaneous abortion was 55.6%, which was significantly lower than that in proliferative endometrium (99.9%), secretory endometrium (87. 5% ) and normal early pregnancy (86.4%) (P 〈0. 05). The high expression rates of △Np63 in groups of secretory endometrium and spontaneous abortion were 16. 7% and 11.2% respectively, which were significantly lower than that of proliferative endometrium (63.6%) and that of normal early pregnancy (57.6%) ( P 〈 0. 01 ). Conclusion △Np63 might take part in the periodic physiological change of human endometrium, and is closely associated with the transformation from human endometrium to decidua of early pregnancy as well as the maintenance of early pregnancy.
