摘要
以淀粉加工副产物玉米胚为原料,经过超临界流体萃取脱脂及超声波破壁处理后,采用热水浸提法提取谷胱甘肽,最佳热水浸提工艺条件为固液比1∶15 g/mL、浸提温度90℃、浸提液pH3.0、浸提时间15 min,在此条件下谷胱甘肽提取量为97 mg/100g玉米胚。选用PES膜进行超滤,确定超滤的最佳膜孔径为0.1μm,最佳超滤压力范围为0.1-0.15 MPa,蛋白质截留率为69.76%,GSH回收率为90.35%。经过对比试验采用732阳离子交换树脂进行谷胱甘肽粗提液的纯化,洗脱液选择浓度为0.2 mol/L的NaOH溶液,谷胱甘肽最大回收率为68.37%。
With corn embryo being coproduct in corn starch production as material, glutathione(GSH) was extracted through hot water extraction after supercritical CO2 fluid extraction and ultrasound wave treatment. The results showed that The optimum conditions of hot water extraction are solid:liquid 1:15, temperature 90 ℃, pH3.0, time is 15 min. On these conditions GSH yield is 97 mg/100 g corn embryo. The PES membrane is selected in this research to carryon ultra filters, and determined the best membrane aperture is 0.1 μm, the best ultrafiltersthe pressure scope is: 0.1-0.15 MPa, protein temporarily stopping rate is 69.76%, the GSH returns-ratio is 90.35%. After the contrast experiment, the 732 cation resin is used to depurate the GSH solution, the concentration of the eluant is the 0.2 mol/L NaOH solution, the GSH maximum returns-ratio is 68.37%.
