摘要
目的:探讨宫颈鳞状上皮内瘤变(CIN)、宫颈鳞癌组织中Skp2和p27的表达及相关性。方法:免疫组化二步法检测60例宫颈鳞状上皮内瘤变(CINⅠ、CINⅡ、CINⅢ各20例)、50例宫颈浸润癌中Skp2和p27的表达,20例正常宫颈组织做对照。结果:S期激酶相关蛋白2(S-phase Kinase-associated Protein 2,Skp2)促进泛素介导的细胞周期素依赖激酶抑制剂p27蛋白降解,研究发现Skp2过表达参与细胞转化和肿瘤的形成。①Skp2蛋白表达CINⅡ明显高于CINⅠ和正常宫颈组织(P<0.05);CINⅠ和正常宫颈组织中差异无统计学意义(P>0.05);CINⅢ明显高于CINⅡ(P<0.05);宫颈浸润鳞癌明显高于CINⅢ(P<0.05)。p27蛋白表达CINⅠ与在正常宫颈组织中差异无统计学意义(P>0.05),CINⅡ明显低于正常宫颈组织和CINⅠ(P<0.05),CINⅢ明显低于CINⅡ(P<0.01)。宫颈浸润癌明显低于CINⅢ(P<0.01)。②宫颈癌Skp2与p27蛋白表达呈负相关。③宫颈浸润癌的Skp2蛋白表达和分化程度、淋巴结转移、临床分期呈正相关。结论:Skp2和p27蛋白可能参与宫颈癌的发生及转移,并与分化程度和临床分期有关。检测Skp2和p27蛋白表达对早期诊断宫颈鳞癌、确定肿瘤恶性程度及预后有重要作用。
Objective: Mainly to investigate the different expressions of Skp2 and p27 in cervical intraepithe-lial neoplasia (CIN) and squamous cell carcinomas of the cervix and the correlation of their expressions with the clinic pathological types. Methods: 50 cases with squamous cell carcinomas of the cervix, 60 cases with cervical intraepithelial neoplasia (CIN Ⅰ , CIN Ⅱ and CINⅢ, n = 20 respectively), 20 cases normal women as control group. All simples were detected by immunohistochemical assay with two steps. Results: Skp2 can accelerate the degradation of p27 whose biological function depends on ubiquitin. Many studies demonstrated that excessive expression of Skp2 plays an important role in cell transformation of malignant tumor.①The expression of Skp2 protein in CIN Ⅱ group was significantly higher than that in CIN I group and normal control group(P〈0.05), the expression in CINⅢ group was higher than CIN Ⅱ group (P〈0.05) , the invasive squamous cell carcino- real of cervix was significantly higher than the CIN Ⅲ group(P〈 0.05), there was no significant difference be-tween the CIN I group and the normal control group(P 〉 0.05). The expression of p27 protein has no significant difference between the CIN I group and the normal control group ; however, the expressions in the CIN Ⅱ group were significantly lower than the normal control group and the CIN I group (P 〈 0.05) ; the expressions in CIN Ⅲ group were significantly lower than the CIN Ⅱ group (P 〈 0.01);the invasive carcinoma of cervix group was distinctly lower than the CIN Ⅲ group(P 〈 0.01 ). ②The expression of Skp2 was negatively correlated with the expression of p27.③The expression of Skp2 in squamous cell carcinomas group was positively correlated with the degree of differentiation, lymphatic metastasis and the clinic pathological type in cervix cancer. Conclu- sion: Skp2 and p27 protein possibly participate in the occurence and metastasis of cervical carcinomas, and they have correlation with the degree of differentiation and the clinic pathological type. Therefore, the detection of Skp2 and p27 is important for the early diagnosis, confirmation of malignant degree and the prediction of cervix cancer.
