摘要
目的筛选以井冈霉素为底物,转化产生井冈霉胺的菌株。方法通过以井冈霉素为碳源,富集培养能够利用井冈霉素的菌株,划线分离出单菌落。各种菌株转化井冈霉素后的产物经衍生化反应后以H PLC法分析。结果经过对225株细菌、真菌和放线菌的筛选,最终得到一株以井冈霉素为底物裂解产生井冈霉胺的菌株Y07B-0134。结论经形态学、生理生化鉴定和16S rDNA基因序列对比分析,确定该菌株为多食鞘氨醇杆菌(Sphingobacterium multivorum)。
Objective To screen a strain which is able to transform validamycin to validamine. Methods Validamycin used as the only substrate culture, using streak culture to pick out the single colony which could transform validamycin to validamine through HPLC analysis. Results Strain Y07B-0134 was screened out as the positive bacterium from 225 strains. Conclusion Based on the analysis of morphological, physiological and biochemical characteristics and the Blast analysis of 16S rDNA, strain Y07B-0134 was identified as Sphingobacterium multivorum.
