SD大鼠重型创伤性颅脑损伤后皮质Shank1a蛋白的改变及意义

摘要 目的探讨Sprague-Dawley(SD)大鼠重型颅脑损伤后皮质组织中Shank1a蛋白的表达改变及其意义。方法48只成年雄性SD大鼠,按随机数字表法,分为正常对照组、颅脑损伤后1h、6h、24h、48h及72h共6组,每组8只。采用免疫组化染色法和Western blot法行蛋白测定,RT-PCR法行mRNA测定。观察各颅脑损伤组及对照组大鼠Shank1a蛋白及mRNA梯度灰度值,并进行免疫组化评分。结果与对照组比较,严重颅脑损伤后,Shank1a蛋白和mRNA的表达量增加,从伤后1h持续到伤后72h(P<0.05),伤后24h表达量最高(P<0.01)。结论在严重颅脑损伤后,Shank1a出现1个表达高峰,在不同机制的影响下可能对脑损伤发生、发展起重要作用。 Objective To investigate the expression and significance of Shankla protein after severe traumatic brain injury in SD rats. Methods Forty-eight adult SD rats were divided into 6 groups with 8 rats for each group: normal control group and post-traumatic groups which respectively are 1, 6, 24, 48 and 72 h groups. Shankla protein was detected by immunohistochemistry and Western blotting and the mRNA by RT-PCR. The protein and gradient gray scale ofShankl a in each group were observed and graded by immunohistochemistry Results Compared with control group, the level of protein and mRNA of Shankla increased gradually from 1h to 72 h after severe traumatic brain injury (P〈0.05), and they reached the peak at 24 h (P〈0.01). Conclusion After sever traumatic brain injury, there is one peak level of Shankla expression and the increased level may play important role in the development and progression of brain injury with different potential mechanism.
