内皮素1对大肠癌HCT-116细胞增殖的影响

Effects of endothelin-1 on the proliferation of human HCT-116 cells

目的探讨内皮素1对大肠癌HCT-116细胞增殖的影响。方法应用RT-PCR及免疫组织化学法检测大肠癌HCT-116细胞内皮素1，内皮素受体A、B表达情况；应用MTT法检测不同浓度和作用时间的内皮素1对体外培养大肠癌HCT-116细胞增殖的影响，多组比较采用方差分析。结果HCT-116细胞存在内皮素l及内皮素受体A、B的基因和蛋白表达；内皮素1能明显促进大肠癌HCT-116细胞的增殖，在干预48h且浓度为1×10^-7mol／L时达到最强。结论内皮素1通过细胞膜上受体的介导，促进HCT-116细胞的增殖，呈药物浓度依赖性。

Objective To investigate the effects of endothelin-1 on the proliferation of HCT-116 cells. Methods The expression of endothelin-1 and endothelin receptors A and B in the HCT-116 cells was detected by RT-PCR and immunohistochemistry method. The effects of endothelin-1 at different concentrations and action time points on HCT-116 cells were measured by MTF assay and were analyzed by variance analysis. Results The expression of endothelin-1 and endothelin receptors A and B at the gene and protein levels in HCT-116 cells was detected. Endothelin-1 had significant effects on the proliferation of HCT-116 cells. The proliferation peak of HCT-116 cells was reached when the concentration of endothelin-I was 1×10^-7 mol/L and the action time was 48 hours. Conclusions Endothelin-1 promotes proliferation of HCT-116 cells in a dose- and time-dependent manner, and the process is mediated by endothetin receptors.
