硒酸精氨酸对D-半乳糖诱导的衰老小鼠肝损伤的保护作用 被引量:3

摘要 目的探讨硒酸精氨酸对D-半乳糖(D-gal)衰老小鼠肝功能的影响。方法40只昆明种小鼠随机分成4组:正常对照组、衰老模型组、低硒组和高硒组。通过后颈背部皮下注射D-gal建立亚急性衰老模型,以灌胃方式加入硒酸精氨酸,连续处理6w后检测各组小鼠血清中谷丙转氨酶(ALT),谷草转氨酶(AST)活性和碱性磷酸酶(AKP)活性,并对肝脏切片做HE染色,光镜下观察肝组织病理学变化。结果硒酸精氨酸能显著降低AST、ALT和AKP活性(P<0.01),明显减轻D-gal对小鼠肝细胞的毒性。结论硒酸精氨酸对D-gal所致衰老小鼠的肝功能具有明显的保护作用,这种保护效应可能与硒酸精氨酸提高抗氧化酶活性,增强抗氧化防御机制有关。 Objective To explore the effect of seleno-argnine on hepatic function of D-galactose (D-gal) induced aging mice. Methods Mice were randomly divided into four groups (n = 10 ) :normal, aging model, low (8.35μg·kg^-1· d^-1 )and high dose seleno- argnine groups( 16. 78 μg·kg^-1· d^-1). The seleno-argnine was added by intragastric administration. Aging model mice were induced by subcutaneous injection D-gal. After 6 weeks of treatment, the level of ALT, AST and AKP in serum were measured. Liver sections were stained with HE, and observed under the microscope. Results Seleno-argnine injection could significantly decrease the activities of AST, ALT and AKP in serum of aging model mice, and reduce the toxic effect of D-gal on liver cells. Conclusions Seleno-argnine has protective effect on D-galactose induced hepatic injury in aging model mice by decreasing the activity of antioxidase and enhancing antioxidant defenses.
