摘要 目的探讨肾上腺髓质素(ADM)在糖尿病大鼠肾脏的表达以及氯沙坦对其的干预作用。方法单侧肾切除大鼠腹腔注射链脲佐菌素(STZ)诱发实验性糖尿病模型,随机分为对照组、糖尿病组和治疗组,治疗组每日用氯沙坦灌胃共6w。采用免疫组化检测ADM和肾上腺髓质素受体(ADMR)的表达,Western印迹检测肾皮质ADMR、转化生长因子β1(TGFβ1)和Ⅳ型胶原(ColⅣ)蛋白的表达,原位杂交检测ADM mRNA的表达。结果各指标糖尿病组较对照组表达均显著增强,治疗组较糖尿病组表达均减低(P<0.01)。结论提示ADM可能在糖尿病早期发挥对肾脏的保护作用,氯沙坦可能部分通过影响ADM及其受体而发挥作用。 Objective To investigate the expression of adrenomedulin (ADM) in the kidney of diabetic rats and the therapeutic effect of losartan. Methods Experimental diabetes models were induced in uninephrectomized Wistar rats by intraperitoneal injection of STZ. The rats were divided into control, diabetic and treatment groups. Treatment group rats were lavaged with losartan. The rats were respectively sacrificed at 6 weeks after injected. The expressions of ADM and ADM receptor (ADMR) were measured by immunohistochemistry, the expressions of ADMR, TGFβ1 and ColⅣ protein were tested by Western blot and the expression of ADM mRNA was tested by hybridization in situ. Results Compared with control group, their expressions in diabetic group were significantly increased, and those in treatment group were lower than those in diabetic group. Conclusions ADM has a protective effect on renal at early stage of diabetes, and losartan may play a role through ADM and ADMR.
