神经生长因子微球对痴呆鼠行为学的影响

摘要 目的观察神经生长因子(rhNGF)微球对老年性痴呆(AD)模型鼠学习和记忆能力的影响。方法采用双乳化技术制备rhNGF微球;雄性SD大鼠28只,随机分为正常组(8只)、模型组(8只)和rhNGF微球治疗组(rhNGF组)(12只)。切断大鼠左侧穹隆海马伞,随即行基底前脑注射rhNGF微球;4w后,Y-迷宫检测各组大鼠的学习和记忆能力。结果模型组大鼠的学习记忆能力明显下降,学习次数和正确记忆次数分别为98.33±8.78和4.0±0.56;rhNGF组大鼠的学习记忆能力得到明显的改善,学习次数显著下降(68.86±4.91,P<0.01),正确记忆次数显著增加(6.8±0.86,P<0.01)。结论rhNGF微球移植治疗能够明显提高AD模型鼠的学习记忆能力。 Objective To study the effect of implantation of recombinant human nerve growth factor (rhNGF) microspheres on the abilities of learning and memory of rats with Alzheimer's disease (AD). Methods rhNGF microspheres were prepared by a W/O/W emul- sion technology. Twenty-eight male SD rats were randomly divided'into normal ( n = 8), model ( n = 8 ) and rhNGF treated group ( n = 12). rhNGF microspheres were transplanted into the basal forebrain after fornix-fimbria transactions. After 4 weeks, the abilities of learning and memory in the different groups were observed by Y-maze. Results After 4 weeks, the abilities of learning and memory of model group were significantly impaired. The times of learning and memory were 98. 33±8.78 and 4. 0 ±0. 56 respectively. In rhNGF treated group, the abilities of learning and memory were significantly improved. The times of learning and memory were 68.86 ± 4. 91 and 6. 8±0. 86 ( all P 〈 O. 01 ) respectively. Conclusions rhNGF microspheres implanted into basal forebrain can improve the learning and memory of the rats with AD.
