胃蛋白酶-胰蛋白酶体外消化豆粕适宜条件的研究 被引量:16

摘要 本试验采用单因素试验设计,在恒温摇床中,用胃蛋白酶和胰蛋白酶体外酶解豆粕,从而确定胃蛋白酶-胰蛋白酶两步法评定蛋白质的体外消化的最适条件。结果表明,优化后的胃蛋白酶-胰蛋白酶两步酶解处理豆粕的主要参数是:温度为37℃,转速85r/min,底物的量为0.5g,缓冲液pH为1.2,胃蛋白酶酶浓度为50mg/mL,锥形瓶中消化6h后,调节消化液的pH至7.5,添加170mg/mL胰蛋白酶溶液,在39℃恒温摇床中培养18h。 A single-factor test design and the homeothermal rocking bed were taken to study the optimum condition of soybean which was digested by pepsase-parenzyme in vitro.The results showed that the optimum condition was that the temperature was 37℃ ,the rate of rotation was 85 r/min,the amount of substrate was 0.5 g,the pH of buffer was 1.2,the concentration of pepsase was 50 mg/mL.After 6 h digestion in conical beaker,the pH value was adjusted to 7.5,170 mg parenzyme was added and cultured 18 h in 39 ℃ homeothermal rocking bed.
