组织芯片技术检测Galectin-3和CD44v6在子宫内膜癌中的表达及其意义 被引量:5

摘要 目的:研究Galectin-3和CD44v6在子宫内膜癌发生、发展中的意义。方法:应用免疫组化ElivisionTM法检测Galectin-3与CD44v6在18例正常增生(NE)、22例简单-复杂型增生过长(SH-CH)、30例不典型增生(AH)子宫内膜及58例子宫内膜癌(UEC)中的表达。结果:①Galectin-3、CD44v6在NE、SH-CH、AH和UEC中的表达逐渐升高,差异有显著性(P<0.05);②Galectin-3表达强度在UEC不同病理分级之间、淋巴结转移与否及肌层浸润深浅差异均有显著性(P<0.05),与临床分期及年龄不相关(P>0.05);③CD44v6表达在UEC临床分期之间、淋巴结转移与否均有显著性差异(P<0.05),与分化程度、肌层浸润及年龄均不相关(P>0.05);④Galectin-3与CD44v6表达在UEC中呈显著正相关。结论:①Galectin-3的过度表达与子宫内膜异常增生及恶变有关;②CD44v6的阳性表达可以用来预测子宫内膜癌淋巴结转移状况以及对恶性程度的判断;③Galectin-3及CD44v6的共同表达有助于尽早发现子宫内膜癌的癌前病变及鉴别子宫内膜的良恶性病变。 Objective: To study the role of Galectin-3 and CD44v6 in the pathogenesis mechanism of endometrial carcinoma. Methods: Immunohistochemical staining (EliVision) was used to detect the proteins of Galectin-3 and CD44v6 in 18 cases of normal endometrium (NE), 22 cases of simple-complex endometrial hyperplasias (SH-CH), 30 cases of atypical endometrial hyperplasias (AH) and 58 cases of uterine endometrioid adenocracinomas (UEC). Results:①The positive expression rate of Galectin-3 and CD44v6 in NE, SH-CH, AH and UEC increased progressively, and the difference was statistically significance (P〈0.05). ② The expression intensity of Galectin-3 was related with the differentiation degree, with/without lymphatic metastasis or muscular layer invasive parameter of UEC (P〈0.05). There were no statistical significances in operation staging and age of UEC. ③The expression intensity of CD44v6 was related with the operation staging and with/without lymphatic metastasis (P〈0.01). There was no statistical significance of CD44v6 expression in differentiation degree, muscular layer invasion or age parameter of UEC. ④The positive expression rate of Galectin-3 was correlated with that of CD44v6. Conlusions:① The overexpression of Galectin-3 reflects the malignant level and invasive ability of endometrial carcinoma indirectly; ② The overexpression of CD44v6 may predict the status of invasion.lymph node metastasis and prognoses in endometrial carcinoma. ③The expression of galectin-3 and CD44v6 plays important roles in the process of the early detection of precancerous lesion and is important in the early diagnosis of endometrial carcinoma.
