摘要
采用组织破碎直接培养法在体外建立一株人低分化原发性肝癌细胞系TJ3ZX-01,并对其生物学特征进行鉴定,利用免疫细胞化学染色与流式细胞术分析,探讨低分化原发性肝癌起源。细胞染色结果表明CK18+、CK19+、波形蛋白+、AFPlow、PCNA+、Hep-,认为该细胞系具有上皮细胞的特征,但肝细胞标记物表达较低,不具有明显的成熟肝细胞特征;利用流式细胞仪分析细胞表面标记物CD29、CD34、CD45、Thy-1(CD90)、CD133、CD49f在细胞中的表达情况,其阳性细胞的比例在传代过程中保持稳定的水平,其中少量细胞可能具有干细胞与前体细胞的性质,显示此例低分化原发性肝癌的来源为肝干细胞或前体细胞。
Cancer cell line TJ3ZX-01 were separated from poorly differentiated hepatocellular carcinomas specimen by the tissue dissection method, and their biological characteristics were identified. The cellular origin of the poorly differentiated HCC cell line was investigated with immunocytochemistry and flow cytometry analysis. Immunocytochemistry detection showed CK18+, CK19+, vimentin+, AFPlow, PCNA+ and Hep, which demonstrated the cancer cell line had the characteristics of epithelial cells but not the obvious characteristics of liver cells for the markers of the liver cell were low, The liver cell and stem cells surface markers CD29, CD34, CD45, Thy-1 (CD90), CD133 and CD49f expressed in the cell line were stable with passages. A small subset of cells in the cancer cells migth has the characters of stem cells or progenitor cells, which indicated that this hepatocellular carcinomas cell line originated from stem cells or progenitor cells.
