DHPLC法分析甘肃东乡族RBP4基因多态性与超重、肥胖、胰岛素抵抗的相关性

摘要 目的探讨甘肃东乡族视黄醇结合蛋白4(RBP4)基因多态性与超重肥胖、胰岛素抵抗的关系。方法用DHPLC技术筛查东乡族113例正常体重者及107例超重肥胖者RBP4基因第五内含子+7542位点的多态性。结果 (1)DM组+7542位点基因型和等位基因频率分布在体重正常和超重肥胖者之间无统计学差异。非DM组超重肥胖者TT基因型和T等位基因频率明显高于体重正常者。(2)在非DM人群中超重和体重正常组均发现TT基因型者的平均HOMA-IR值明显高于其他基因型者。结论甘肃东乡族RBP4基因多态性可能与超重肥胖、胰岛素抵抗相关。 Objective To study the association of RBP4 gene polymorphism with overweight, obesity and insulin resistance in Dongxiang people of Gansu province. Methods The single nucleotide polymorphism of +7542 locus in RBP4 was detected by denatured HPLC in 113 subjects characterized as normal weight and 107 subjects characterized as overweight and obesity. Results (1) The distribution of genotype and allele frequency had no significant difference between the normal weight subjects and overweight/obese subjects in type 2 diabetes group. The frequencies of TT genotype and T allele were significantly higher in the overweight/obese subjects than in the normal weight subjects in non-diabetes group (P= 0. 006,0. 008). (2) The insulin resistance index (HOMA-IR) of TT genotype was higher than of the other genotypes both in the normal weight and overweight/obese subjects in non-diabetes. (P= 0. 015,P=0. 022). Conclusions The polymorphism of RBP4 gene may be related to insulin resistance, overweight and obesity in Dongxiang people of Gansu province.
