摘要
目的:建立同时测定维吾尔药材天山花楸中金丝桃苷和芦丁含量的RP-HPLC分析方法。方法:采用HPLC法,色谱柱为Symmetry shield RP8(5μm,4.6 mm×150 mm);流动相为甲醇-0.05%磷酸(三乙胺调pH至3.0)(40∶60),柱温40℃,流速1.3 mL·min-1,检测波长360 nm。结果:金丝桃苷和芦丁分别在20.0~100.0 ng和15.8~79.0 ng范围内,进样量与色谱峰积分面积呈良好线性关系,相关系数分别为0.9997和0.9999,平均回收率(n=6)分别为102.3%和100.5%。结论:应用本法测定金丝桃苷和芦丁的含量稳定可靠,重复性好,可用于天山花揪药材的质量控制。
Objective:To establish RP - HPLC method for the determination of hyperoside and rutin in Sorbus tian-shanica Rupr. from Xinjiang. Method:Symmetry Shield RP8 column (4. 6 mm× 150 mm,5μm) was eluted with mobile phase consisted of methanol and 0. 05% aqueous phosphoric acid (40:60 ,adjusted to pH 3.0 with triethyl- amine) at the column temperature 30 ℃. The flow rate was 1.3 mL · min^-1 and detective wavelength was 360 nm. Results: Hyperoside was proved to be linear in the range of 20.0 - 100. 0 ng with a regression coefficient of 0. 9997 ,the average recovery was 102.3% ;rutin was proved to be linear in the range of 15.8 -79.0 ng with a regression coefficient of 0. 9999, the average recovery was 100.5%. Conclusion: The method is proved to be quick, simple and high sensitive.
