摘要
目的:研究灯盏细辛注射液(Erigeron breviscapusinjection,EBI)对离体培养的大鼠脑微血管内皮细胞(BMEC)炎症相关细胞因子白细胞介素-1β(IL-1β)及细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)蛋白表达的影响,初步探讨EBI抗脑缺血炎症损伤的机制。方法:采用肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)建立离体培养的大鼠BMEC炎症损伤模型,分别用0.1 mg.L-1和1 mg.L-1EBI处理内皮细胞,使用ELISA试剂盒分析IL-1β蛋白表达的变化,流式细胞仪测定ICAM-1和VCAM-1荧光强度的变化。结果:EBI可以抑制TNF-α诱导的内皮细胞IL-1β高表达,降低TNF-α诱导的内皮细胞ICAM-1和VCAM-1高表达。结论:EBI可以降低由TNF-α诱导的内皮细胞过度表达IL-1β,ICAM-1和VCAM-1,从而对脑缺血炎症损伤具有一定保护作用。
Objective: To investigate the effect of Erigeron breviscapus injection (EBI) on the expression of interleukin-1β (IL-1β), intercellular adhesion molecule-1 (ICAM-1), and vascular cell adhesion molecule-1 ( VCAM-1 ) in rat brain microvascular endothelial cells (BMEC) cultured in vitro. Methods : The inflammatory injury was induced by tumor necrosis factor-α (TNF-α) in BMECs. The endothelial cells were treated with EBI at 0. 1 and 1 mg·L^-1 The expression of IL-1β protein was detected by ELISA, and those of ICAM-1 and VCAM-1 were measured by flow cytometry. Results : EBI inhibited TNF-α-enhanced expression of IL-1β in BMECs. Meanwhile it diminished TNF-α-enhanced expression of ICAM-1 and VCAM-1 in BMECs. Conclusion: EBI decreases TNF-α- enhanced expression of IL-1β, ICAM-1 and VCAM-1 in BMECs, resulting in a protective effect on inflammatory injury after cerebral ischemia.
