川芎嗪含药血清干预软骨细胞周期作用机制的研究

Empirical study on the mechanism of action to blood serum contained with chuanxiongzine interventing cartilage cell cycle

目的:探讨川芎嗪含药血清干预软骨细胞周期的作用机制。方法:取4周龄新西兰兔膝关节关节软骨Ⅱ型胶原酶消化,建立软骨细胞体外培养体系,体外培养第三代软骨细胞随机分为3组,分别加入培养液、空白血清、含药血清继续培养24 h、48 h后,采用流式细胞仪检测软骨细胞周期的变化。结果:流式细胞仪细胞周期检测显示川芎嗪含药血清能够刺激软骨细胞由G0/G1期进入S期和G2/M期,并呈现时间的依赖性,干预48 h后G0/G1期细胞比例降低,S期与G2/M期细胞之和增加,与正常组、空白组有显著性差异(P<0.05)。结论:川芎嗪含药血清能有效推动软骨细胞周期的进程。

Objective：To approach the mechanism of action on blood serum contained with chuanxiongzine interventing cartilage cell cycle. Methods ： Newzealand rabbit with four - week age were choosed and their arthrodial cartilage of knee -joints were digested by type II collagenase to make models for building culture system of cartilage cell in vitro. Cartilage cell of the third generation cultured in vitro were randomly divided into three groups, culture fluid, blank serum and serum contained with drug were respectively added into them, and continued to be cultured for 24 hours and 48 hours. Then the changes of cartilage cell cycle were detected by flow cytometry. Results ： The detection of cell cycle by flow cytometry showed that the serum with chuanxiongzine could stimulate cartilage cell from G0/G1 period to S period and G2/M period, the course was temporal dependence. Interventing 48 hours later, the ratio of cells in G0/G1 period degraded, and cell sum in periods of S and G2/M increased, there had significantdifference between it and normal group as well as blank group （ P 〈 0.05 ） . Conclusion： the Serum contained with chuanxiongzine could effectively improve the preceeding of cartilage cell cycle.
