苯乙酸钠对Hela细胞的生长抑制和分化诱导

摘要 研究非毒性分化诱导剂苯乙酸钠(NaPA),在体外对子宫颈癌Hela细胞的生长、分化及表型的影响。实验结果:1.NaPA能够抑制Hela细胞的着壁依赖性和着壁不依赖性生长.而对正常羊膜组织Wish细胞的生长无显著抑制作用;2.NaPA能够增强Hela细胞表面ICAM-1.HLAl以及Ⅱ类分子的表达:3.以电子显微镜观察到在NaPA处理的Hela细胞中,线粒体、内质网以及弹力丝等丰富.而在NaPA未处理的细胞中,除了大量游离的多聚核糖体之外.上述的细胞结构几乎没有观察到。结果表明NaPA在体外不仅能够选择性抑制子宫颈癌细胞的生长,而且能够有效地诱导细胞的分化及表型转变,为NaPA分化治疗子宫颈癌提供了实验依据。 Differentiation therapy which is different from radiotherapy and chemotherapy may provide a new treatment for cancers. This led us to evaluate the effect of a nontoxic differentiation inducer, sodium phenylacetate (NaPA) on cervical cancer Hela cells in vitro. NaPA treatment resulted in dose-dependent inhibition of Hela cell growth, including anchorage-dependent and anchorage-independent growth. But NaPA was not significantly inhibitory to the growth of normal amniotic tissue Wish cells. In addition, NaPA could enhance the expression of ICAM-1, HLA class Ⅰ and Ⅱ molecules on the surface of Hela cells. Electron micrographs of NaPA-treated Hela cells demonstrated the emergence of rich mitochondria, rough endoplasmic reticulum and elastic filament, indicating the active cell differentiation, however, in NaPA-untreated Hela cells, the abovemen tioned structures cannot be observed except numerous freely scattered polyribosomes. These results suggest that NaPA in vitro not only selectively inhibit the growth of cervical cancer cells, but also effectively induce the differentiation and phenotype transformation of these cells, providing experimental basis of NaPA differentiation treatment of cervial cancer.
