骨髓基质干细胞在骨内示踪方法的研究 被引量:1

摘要 目的探讨狗骨髓基质干细胞(MSCs)的骨内示踪技术。方法体外培养、增殖骨髓基质干细胞,并在体外以Brdu、PKH-26标记,将标记好的细胞移植回狗股骨头内,观察细胞的示踪情况。结果Brdu、PKH-26均能有效的标记骨髓基质干细胞,其标记率分别为85%、93%;移植细胞1个月后狗股骨头内可见到明显的Brdu着色的成骨细胞,PKH-26标记法可见股骨头内明显的PKH-26着色,但有染料外渗现象发生。结论Brdu是外源性细胞体内示踪理想的标记方法,PKH-26如能注意要点,也是一种比较有效、实用的标记方法。 Objective To explore a method of tracing technique of MSCs in vivo. Methods Culturing and proliferating MSCs in vitro, using Brdu and PKH-26 to trace MSCs,and then transplanted MSCs into Dog's femoral head, observing MSCs changes in vivo. Results MSCs were labeled well by Brdu and PKH-26, the labeled rates of Brdu and PKH-26 were 85% and 93%. After one month, we can find osteoblasts which were conspicuously suffused by Brdu in Dog' s femoral head. we can also find visible tinction by PKH-26, but which had dye exosmosis phenomenon was also detected. Conclusions Brdu is an ideal tracer technique in vivo for exogenous cell. If we can take notice of main points when using the dye of PKH- 26, it is also an effective tracer technique.
