海洋真菌碱提多糖YCC的结构初步鉴定及其对肾小球系膜细胞增殖的抑制作用 被引量:1

摘要 文章对海洋真菌碱提多糖YCC进行了初步结构鉴定,并探讨了其对人肾小球系膜细胞(MCs)增殖的作用。采用了气相色谱法、红外光谱法、高碘酸氧化法、Smith降解等方法测定了多糖的结构。用四甲基偶氮唑盐(MTT)法检测,观察了YCC对高糖培养的MCs增殖的影响。结果表明,YCC的平均相对分子质量为66k,单糖组成为:甘露糖、葡萄糖、半乳糖,其摩尔比为1.8∶81.33∶16.87;比旋度测定及IR结果表明,YCC为α构型;高碘酸氧化及Smith降解的结果显示,YCC的连接方式主要为1→6连接、1→4连接和1→3连接。YCC浓度在1mg/ml时对高糖所致的MCs增殖有显著的抑制作用。 To investigate the chemical structure of the polysaccharides (YCC) and its effect on proliferation of MCs, several methods such as C-C, IR and MTT analysis were used. In conclusion, YCC has a backbone of α(1→6), α(1→4) and α(1→3). YCC can suppress the proliferation activity of MCs cultured in high glucose.
