摘要
目的以红酵母RY-4为试验菌株,欲提高其合成谷胱甘肽(GSH)的能力。方法在发酵期给予一定刺激,分别采用添加KMnO4溶液和提高渗透压的方式对细胞进行处理。结果在发酵30 h添加1 g/L KMnO4溶液0.6mL,刺激细胞发生应激反应,GSH总量和胞内质量分数分别达到255.27 mg/L和5.51%,与对照相比提高35.1%和35.0%;在发酵24 h加入一定量KCl(在发酵液中的终浓度达20 g/L),提高培养液的渗透压,GSH总量和胞内质量分数分别达到274.74 mg/L和5.95%,与对照相比提高46.5%和47.2%。经过试验,将两者作用叠加,GSH总量和胞内质量分数分别达到281.95 mg/L和6.22%,比出发株提高了51.0%和52.1%。结论红酵母RY-4摇瓶培养时可以利用提高渗透压与添加氧化刺激的方法提高GSH的产量。
Purpose To improve the production of glutathione from Rhodotorula RY-4 strain, the stimulating effects by osmotic pressure and oxidative stress were used. Methods 1 g/L KMnO4 was added at 30 h and KC1 20 g/L was added at 24 h. Results When 1 g/L KMnO4 0.6 mL was added at 30 h, the GSH production and intracellular content were improved by 35.1% and 35.0% respectively. And when 20 g/L KC1 was added at 24 h, the GSH production and intracellular content were improved by 46.5 % and 47.2 % respectively. The results also showed that interaction caused by both osmotic pressure and oxidative stress could improve the GSH production and intracellular content by 51.0 % and 52.1%. Conclusion Osmotic pressure and oxidative stress can be used to improve glutathione production effectively.
