摘要
[目的]筛选最佳的金线莲增殖培养基。[方法]以金线莲组培苗的茎段和顶芽为外植体进行增殖培养,在光照时间12 h/d,光照强度1 200 lx,温度(25±2)℃,pH值为5.8培养条件下比较不同基本培养基、细胞分裂素、生长素、添加物等因素对金线莲生长状况的影响。[结果]通过对不同培养基的培养效果进行对比分析后筛选出适宜的培养基分别是:生芽增殖最佳配方为MS+琼脂7g/L+蔗糖25 g/L+NAA 0.2 mg/L+6-BA 2 mg/L;壮苗培养基为MS+琼脂7 g/L+蔗糖25 g/L+蛋白胨2 g/L+花宝1号3 g/L+香蕉100 g/L+活性炭1 g/L和1/2 MS+琼脂7 g/L+蔗糖25 g/L+蛋白胨2 g/L+香蕉100 g/L+活性炭1 g/L。[结论]添加马铃薯、香蕉、蛋白胨、花宝一号、活性炭等添加物能显著地促进金线莲外植体的分化与生长。
[ Objective] The study aimed to screen the optimum multiplication medium of tissue culture for Anoectochus roxburghih. [ Method] The stem segment and terminal bud of the tissue culture seedlings of A. roxburghih were taken as the explant for multiplication culture to compare the effects of factors such as different basic culture medium, cytokinin, growth hormone, supplement matter on the growth of A. roxburghih under the culture conditions as light time of 12 h/d, light intensity of 1 200 lx and temperature of (25 ±2 ) ℃ and pH orS. 8. [ Result ] After the comparative analysis on the culture effect of different media, the screened optimum media were as follows: the optimum formula for budding multiplication was MS + agar 7 g/L + sucrose 25 g/L + NAA 0.2 mg/L + 6- BA 2 mg/L; the strong sprout media were MS + agar 7 g/L + sucrose 25 g/L + protein peptone 2 g/L + Huabao No. 1 3 g/L + banana 100 g/L + activated charcoal 1 g/L and 1/2MS + agar 7 g/L + sucrose 25 g/L + protein peptone 2 g/L + banana 100 g/L + activated charcoal 1 g/L. [Conclusion] The supplement matter such as potato, banana, protein peptone, Huabao No. 1 and activated charcoal could improve the differentiation and growth of A. roxburghih explant.
