摘要
[目的]为了解犬附红细胞体抗原特性。[方法]对犬附红细胞体进行体外培养,培养后对解离的犬附红细胞体经超声波裂解,制备粗提抗原并提纯,对所提抗原进行SDS-PAGE及免疫印迹分析。[结果]犬附红细胞体抗原在电泳图谱上出现了3条主蛋白带,其分子量分别为66、32和25 kD,其中32和25 kD的抗原具有较好的免疫性,且不与犬附红细胞体阴性血清发生交叉反应,具有较好的特异性。[结论]该研究为犬附红细胞体的抗原成分及犬附红细胞体病的免疫学诊断等研究奠定了基础。
[ Objective ] This study aimed to grasp antigen traits of Eperythrozoon canis. [ Method] After culturing Eperythrozoon canis in vitro and splitting the dissociated Eperythrozoon canis with ultrasonic wave, this study analyzed the extracted antigen by SDS-PAGE and immunoblotting. [ Result] 3 main protein belts appeared on the electrophoretogram of Eperythrozoon canis, their molecular weight were respectively 66, 32 and 25 KD, among which antigen of 32 and 25 KD had better histocompatibility and would not cross react with negative serum of Eperythrozoon canis. [ Conclusion ] This research provided foundations for the antigen of Eperythrozoon canis and immunological diagnosis of Eperythrozoon canis diseases.
