牡蛎不同提取方法中糖原含量的比较 被引量:10

摘要 目的通过含量测定,筛选适宜的牡蛎糖原提取方法。方法采用比色法,以苯酚-硫酸溶液为显色体系,检测波长为490nm。结果葡萄糖进样量线性范围为10~80μg(r=0.9988),平均回收率97.74%,RSD=0.91%(n=6)。水提法获得的样品糖原含量最高,为48.46%;经SephadexG-50凝胶柱层析收集第Ⅱ峰,其冻干品的糖原平均含量为26.12%。结论比色法方法简便、准确、重现性好,可用于牡蛎中糖原含量测定。 Objective To set up an assay method for the determination of glycogen in oyster, and to chose the eligible abstraction method. Methods The samples were determined by spectrophotometry with phenolsulfuric acid solution as coloring reagent. The detective wavelength was 490 nm. Results The calibration curve was linear in the range of 10-80μg and the average recovery rate was 97.74%, RSD was 0.91% (n =6). Glycogen measured from water extraction reached to the highest levels 48.46%; in the second peak collected by the gel chromatography columns Sephadex G-50, the average levels of glycogen of the freeze-dried goods was 26. 12%. Conclusion The method is reliable and accurate with good reproducibility. It can be used for the quality control of glycogen in ovster.
