生防菌枯草芽孢杆菌BS-5在3L发酵罐中的发酵工艺优化 被引量:9

摘要 为了提高生防菌枯草芽孢杆菌BS-5液体发酵的生物量,在3L发酵罐中进行了分批发酵和补料发酵试验。在分批发酵中,将氮源物质豆粕浓度由2%提高到4%之后,生物量由4.8×109cfu/ml提高到8.2×109cfu/ml。在分批补料发酵中,采取分批多次补糖策略。初糖浓度为20g/L,当糖浓度在10g/L以下时,补加葡萄糖(每次补加量为20g/L),共补加两次,生物量提高到2.8×1010cfu/ml。 In order to increase the biomass of Bacillus subtilis, batch fermentation and fed-batch fermentation experiments were performed in 3 liter fermentor. Results showed that the biomass was increased from 4.8×10^9 cfu/ml to 8.2×10^9 cfu/ml while the soy bean powder concentration was increased from 2% to 4% at batch fermentation. In the course of fed-batch culture, three modes of glucose feeding strategy were employed, of which the best mode was that the suitable initial concentration of glucose was 20 g/L, when the glucose concentration in broth was below 10 g/L, condensed glucose was fed into the broth twice, the supplying quantity was 20 g/L every time, the biomass was up to 2.8×10^10 cfu/ml.
