空间搭载高产葡萄糖耐量因子(GTF)酵母的选育 被引量:4

Selection of a High Yield Yeast Strain of Glucose Tolerance Factor After Space Piggyback Experiment

摘要 以从本试验室收藏的7株酵母菌种及其7株经空间搭载(实践八号)诱变后的酵母菌株为研究对象,通过梯度含铬YEPD平板培养和液体YPD培养基发酵试验,利用氨水提取菌体中的GTF和火焰原子吸收光谱法检测其有机铬含量,从中筛选出一株葡萄糖耐量因子的高产酵母(YS-3),有机铬含量达1296μg/g,总铬含量达1926μg/g,生物量为40g/L。同时分析了菌体发酵过程中,菌体有机铬富集量与其它发酵参数的动态关系。 A strain of high-biomass, producing glucose tolerance factor yeast strain was screened from seven yeast strains and seven strains mutated by spaceflight-induced mutagenesis through gradient concentration of chromium plates cultivation and liquid fermentation cultivation. The biomass of it was 40 g/L. Extracting GTF with ammonia and determining the content of Cr with flame atomic absorption spectrometry, the content of organic Cr was 1287 μg/g and that of total Cr was 1917 μg/g. At the same time, dynamic relationship between other parameters and the amount of organic chromium was analyzed during fermentation.
