基质金属蛋白酶-3在原发性干燥综合征患者唇腺中的表达

摘要 目的:研究基质金属蛋白酶-3(matrix metalloproteinases-3,MMP-3)在原发性干燥综合征(primarySjogren′s syndrome,pSS)患者唇腺组织中的表达情况。方法:收集24例pSS患者和14例对照组的唇腺标本,提取唇腺中总RNA,利用反转录聚合酶链反应(RT-PCR)法扩增MMP-3、β-肌动蛋白(β-actin)的片段,计算MMP-3/β-actin吸光度比值。免疫组织化学染色方法检测MMP-3蛋白在唇腺中的表达情况。结果:pSS组唇腺中MMP-3的mRNA转录及蛋白表达水平均高于对照组(P<0.05)。MMP-3蛋白主要分布在腺体导管上皮细胞胞浆内。结论:pSS患者唇腺中MMP-3表达增加,可能是唇腺结构破坏的原因之一。 Objective: To investigate the mRNA transcription and protein expression of MMP-3. Possible roles of MMP-3 in primary sjogren's syndrome(pSS) were analyzed. Methods: For this study, 24 patients with pSS were selected and diagnosed by the American-European Consensus Group standards. The control group was composed of 14 subjects. Total RNA from labial glands was extracted. The target genes and β-actin acted as templates to perform reverse transcript polymerase chain reaction (RT-PCR). Then mRNA expression of target genes was semi-quantitated by the OD ratio of the target genes to β-actin bands on gel under densitometry. Immunohistochemical analysis was made to determine the expression of proteins of targets genes in labial glands of pSS patients and control group. Results: In contrast to control group, the mRNA transcription and protein level of target genes in labial glands of pSS patients were significantly increased statistically ( P 〈 0.05 ), and MMP-3 protein was mainly expressed in intracytoplasm of gland duct. Conclusion: The increase of MMP-3 in pSS patients provides a rationale for understanding the organization observed in the basal lamina and apical surface of acini in these patients.
