山西汉族Y-STR基因座DYS552遗传多态性调查

摘要 目的:调查Y染色体上的短串联重复序列DYS552基因座在山西汉族男性人群中的基因频率分布、多态性及群体分布情况,为法医学个体识别及亲子鉴定提供计算数据,对其在法医学上的应用进行可行性探讨。方法:随机抽取102例山西汉族男性无关个体静脉血500uL,用GDB中的引物进行PCR扩增,银染显色,对其选取的等位基因测序,构建等位基因分型标准物;按照ISFG原则命名各等位基因。结果:102例山西汉族人群DYS552基因座共检出7种不同的等位基因,分别为DYS552×23(0.0194)、DYS552×24(0.1105)、DYS552×25(0.3981)、DYS552×26(0.2427)、DYS552×27(0.1262)、DYS552×28(0.0874)、DYS552×29(0.0097),基因多样性为0.7523,其个人识别能力(DP)和非父排出率(PE)均为0.7523。结论:DYS552基因座具有较高的遗传多态性,在法医学及人类遗传学方面具有较高应用价值。 Objective :To provide the evidence for the application of DYS552 in forensic science and population genetics by investigating its polymorphism in the Han populmion lived in Shanxi province. Methods :To collect respectively 102 unrelated male's venous blood 500 uL,who live in Shanxi province. The products of PCR were analyzed with polyacrylamide gel electrophoresis. Each allele of this Y-STR loci was sequenced and the allele ladders were constructed. Alleles of this Y-STR loci were nominated according to recommendations of the International Society of Forelnsic Genetics. Results:There are total 7 alleles in DYS552 site in Shanxi Han population,called DYS552× 23, DYS552 ×24.DYS552 ×25.DYS552 × 26.DYS552 × 27,DYS552 × 28,DYS552 × 29 ,the GD value is 0. 752 3. If used in forensic science,both of the power of diacrimination(PD)and the power of exclusion(PE)were 0. 7523. Conclusion: DYS552 has a high level of genetic polymorphism and can be used in forensic science and human genetics research.
