原花青素对2型糖尿病局灶性脑缺血大鼠海马组织nNOS的影响

Effect of Procyanidin on Expression of nNOS in Type 2 Diabetes Mellitus Rats with Focal Cerebral Ischemia

摘要 目的探讨原花青素(procyanidin,PC)对2型糖尿病局灶性脑缺血大鼠脑组织中神经元型一氧化氮合酶(nNOS)表达的影响。方法将66只大鼠随机分为假手术组、2型糖尿病单纯脑缺血组及PC组(100 mg/kg),分别于术后3、6、12、24、48 h应用免疫组织化学方法行nNOS半定量分析。结果脑缺血后海马区有大量nNOS免疫反应阳性细胞表达,缺血后3 h开始表达,6 h达高峰,12 h后逐渐降低;在相同的时间点,模型组较假手术组明显升高(P<0.01),PC组较模型组明显降低(P<0.01),与假手术组无明显差异(P>0.05)。结论PC具有降低2型糖尿病局灶性脑缺血大鼠脑缺血海马组织nNOS活性的作用。 Objective To investigate effects of proeyanidin ( PC ) on type 2 diabetes meUitus rats with focal cerebral ischemia about neuronal NO synthase in hippocampus tissue. Methods Sixty -six SD rats were divided into sham -operation, model, and PC group. After ischemia for 3 h, 6 h, 12 h, 24 h, 48 h, the expression of nNOS was examined by using immunohistochemical SABC method. Results There was mass expression of nNOS positive - cells in the hippocampus tissue after isehemia. They appeared 3 hours after isehemia and increased progressively, reached the peak 6 hours after ischemia and then gradually decreased. At the same time, the expression of nNOS positive -cells was higher in model group than that in sham operated group (P 〈0. 01 ) . The expression was lower in PC group than that in model group ( P 〈 0.01 ) , but there was no significant difference between the PC group and sham operated group ( P 〉 0. 05 ) . Conclusions PC can reduce the activity of nNOS in hippocampus tissue of the type 2 diabetes melhtus rats with focal cerebral ischemia.
