摘要
目的观察诱导型一氧化氮合酶抑制剂对关节软骨修复组织胶原表达的影响。方法24只新西兰大白兔双侧股骨髁间关节面造成全层软骨缺损。随机抽签均分为对照组、骨形态发生蛋白(bone morphogenetic protein,BMP)组和一氧化氮合酶抑制剂S-甲基异硫脲组(S-methylisothiourea,SMT)组。术后1年处死动物。应用苦味酸-天狼猩红染色检测胶原的分布情况,图像分析技术定量胶原的表达及进行修复组织软骨厚度的测量。应用免疫组织化学技术检测型胶原的表达和分布。结果图像分析显示,1年后SMT组型胶原表达量为65.9%,明显少于BMP组88.5%和对照组94.6%的表达量,型胶原30.7%的表达量明显多于BMP组10.8%和对照组4.5%的表达量。1年后SMT组软骨厚度(1.85±0.56)mm明显高于BMP组(0.67±0.31)mm和对照组(0.24±0.10)mm。SMT组缺损区型胶原含量明显高于BMP组和对照组。结论iNOS抑制剂SMT的应用能明显增加型胶原表达和软骨厚度,对于维持软骨表型有积极意义。
Objective To discuss effect of collagen expression in articular cartilage repaire tissue by inducible nitric oxide synthase inhibitor SMT. Methods Full-thickness defects of cartilage were created in the trochlear groove of twenty-four adult New Zealand White rabbits, eight defects were empty,eight were filled with a collagen fibrin glue impregnated with rhBMP,eight were filled with a collagen fibrin glue impregnated with rhBMP and hypodermic injection with 5 mg/kg-1/12 h-1 SMT. The animals were killed at one-year postperatively. The tissue sections were stained with Sirius-Red against collagen type- I and type- I which analyzed by Quantiment 500. Cartilage thickness was measured by Quantiment 500 too. Results One-year after collagen fibrin glue with rhBMP and with hypodermic injection SMT,collagen type- I expression of repair tissue in SMT group(65.9%) was less than in BMP group(88.5%) and the control (94.6 % ),and collagen type- Ⅱ (30.7 % ) was more than in BMP group (10. 8 % ) and the control (4.5 % ). Cartilage thickness of SMT group(1.85±0. 56) mm was significant higher than BMP group(0.67±0. 31) mm and the control(0. 24 ± 0. 10) mm. Conclusion NOS inhibitor SMT could increase collagen type- I expression and decrease collagen type- Ⅱ expression,as well as make phenotype good.
