摘要
目的:检测缺氧诱导因子1α(HIF-1α)和基质金属蛋白酶9(MMP-9)在结肠癌组织中的表达情况,探讨其在淋巴结转移中的作用和意义。方法:应用免疫组化方法检测78例结肠癌组织HIF-1α和MMP-9表达情况。结果:结肠癌组织中HIF-1α阳性表达率为73.08%,MMP-9在结肠癌组织中阳性表达率为62.82%,HIF-1α的阳性表达与MMP-9的阳性表达密切相关,r=0.550,P<0.01。HIF-1α阳性表达率在有淋巴结转移组(88.24%)显著高于无淋巴结转移组(44.44%),MMP-9阳性表达率在有淋巴结转移组(86.27%)显著高于无淋巴结转移组(29.63%),HIF-1α和MMP-9的阳性表达与结肠癌的组织学分级和Duke分期密切相关。结论:HIF-1α和MMP-9在结肠癌组织中的过表达可能参与了结肠癌的淋巴道转移,HIF-1α可能通过增强MMP-9的表达来提高肿瘤细胞的侵袭性和转移能力。
OBJECTIVE:To investigate the significance of HIF-1α and MMP-9 expression in colonic carcinoma lymphatic metastasis.METHODS:Expression of HIF-1αand MMP-9 was detected in 78 cases of colonic cancer tissue by immunohistochemistry.RESULTS:The positive rate of HIF-1α in colonic carcinoma was 73.08%,and the positive staining for MMP-9 protein was 62.82%.And significant correlation was found between the expression of HIF-1α and MMP-9 in colonic carcinoma(r=0.550,P〈0.01).In lymphnode metastasis positive and negative groups,the positive rates of HIF-1α in the two groups were 88.24% and 44.44% respectively,and there was significant difference between the two groups;the positive rates of MMP-9 in the two groups were 86.27% and 29.63% respectively,and there was significant difference between the two groups.The expressions of HIF-1α,MMP-9 were associated with the histological grade and Duke staging.CONCLUSIONS:Overexpression of the two proteinsmay participate the lymph node metastasis of colonic carcinoma.Overexpression of HIF-1α can stimulate MMP-9 overexpression may be one of the pathways of HIF-1α protein to contribute to the invasion and metastasis in human colonic cancer.
