人成熟卵母细胞冻融的研究

摘要 目的探讨冷冻保护剂中不同蔗糖浓度对人成熟卵母细胞冻融的影响。方法根据冷冻液中蔗糖浓度的不同,将收集到的人成熟卵母细胞分为0.1mol/L组、0.2mol/L组、0.3mol/L组,分别采用程序化慢冻-快融法,比较三组人成熟卵母细胞复苏后的存活情况及其行单精子卵泡浆内注射(ICSI)后受精率、卵裂率和优质胚胎形成率。结果0.1mol/L组、0.2mol/L组、0.3mol/L组解冻后的存活率分别为23.1%,40.8%,59.4%,三组比较差异有统计学意义(P<0.05);而受精率、卵裂率、优质胚胎形成率三组间比较差异无统计学意义(P>0.05)。结论提高蔗糖浓度能明显提高人成熟卵母细胞解冻的存活率。 Objective To explore the effects of different concentrations of sucrose in cryoprotectant solution on the frozen-thawed human mature oocytes. Methods The human mature oocytes were divided into 3 groups according into the concentrations of sucrose in cryoprotectant solution:group A(0.1 mob/L) ,group B(0.2 mob/L) and group C(0. 3 mob/L). The survival rate,fertilization rate, cleavage rate and formation percentage of high quality embryo were analyzed by slow-freezing and quick-thawing method. Results The survival rates were 23.1% ,40.8 % ,59.4% in group A, group B and group C, respectively ( P 〈 0.05 ). But the fertilization rate, cleavage rate and formation percentage of high quality embryo showed no significant difference between 3 groups (P 〉 0. 05 ). Conclusion Rising concentrations of sucrose in the cryoprotectant solution could obviously enhance the survival rate of human mature oocytes.
