维拉帕米在结肠癌光动力学疗法中增效作用研究 被引量:3

摘要 目的:探讨维拉帕米(Ver)增加癌细胞内光敏剂和光动力学疗法(PDT)杀伤作用的效果。方法:在结肠癌细胞培养的基础上,分别采用荧光分光光度法和噻唑蓝(MTT)法分析癌细胞内光敏剂含量和PDT后细胞存活率。结果:Ver可显著增加光敏剂进入癌细胞内的含量,给药后16小时最明显。单独Ver(0.02μg/ml)对癌细胞存活率无明显影响。激光照射后8,16小时,PSD+Ver组细胞存活率与PSD组相比显著降低,32小时细胞存活率接近;加光敏剂后16小时,激光照射,癌细胞存活率较低,可达满意效果。结论:Ver可显著增加癌细胞内光敏剂含量和光动力学杀伤结肠癌细胞能力。 Objective:To investigate the killing effect of photosensitizer and photodynamic therapy(PDT) by adding Verapamil for cancer cells.Methods:On the basis of cell culture,the content of photosensitizer in cancer cells and cell survival after PDT were detected by fluorescence spectrophotometry and MTT assay,respectively.Results:The study showed that the content of photosensitizer in cancer cells was obviously increased by adding the verapamil,particularly in the 16h of postverapamil.But the verapamil alone didn′t influence the cancer cell survival.After 8,16h of laser irradiation,the cancer cell survival was markedly reduced which compared to the single photosensitizer group.After the photosensitizer was given 16h,the laser irradiation was applied.The satisfactory effect was obtained,that is,the cancer cell survival was lower.Conclusion:The study suggests that the verapamil may increase the content of photosensitizer in cancer cells and enhance the killing effect of PDT for colon adenocarcinoma.
