口腔癌TIL的表型及细胞毒活性测定

Analyses on phenotypes and cytotoxicity of TILs from patients with oral cancer

作者在成功地建立了口腔癌TIL的分离、培养方法的基础上,采用单抗致敏的红细胞花环试验和H^3-TDR释放法,对10例口腔癌TIL的表型及功能特征进行了动态分析。结果发现新分离TIL中CD_3^+、CD_4^+、CD_8^+较低,体外培养后,TIL中各亚群均有增加,但以CD_3^+、CD_8^+增加为主;新分离TIL的NK和LAK活性分别为10％和10.5％,随体外培养时间延长,其细胞毒活性增强,在4周时达高峰。NK和LAK活性分别为50％和44.8％。4周后,其活性开始降低。提示口腔癌TIL的临床应用时机为培养4周左右。

With the methods of rosette test and H3 - TDR release assay respectively, We analysed phenotypes and cytotoxicity of TILs obtained from patient with oral cancer. The results showed that,CD3+ CD4+ and CD8+ lymphocytes scattered in the newly isolated TILs, cultured with low - concentration of rIL-2 in vitro,TILs expressed CD3+ and CDg+ predominantly, though CD4+ cells increased significantly. Their cytotoxicity against both NK - sensitive K562 and LAK - sensitive Raji targets were limited before cultivation(only10.0% and 10.5% respectively) .increased along with the time of proliferation and reached the peak(up to 50.0% and 44.8% respectively)at the 4th culture- week. This study demonstrated that the clinical transfer of TILs is administrated after four - week incubation in vitro.
