基质金属蛋白酶抑制因子3甲基化与急性白血病的相关性研究 被引量:1

摘要 目的探讨基质金属蛋白酶抑制因子3(TIMP3)基因启动子区域甲基化在急性白血病发病机制中的作用。方法采用甲基化特异性聚合酶链反应(PCR)法检测50例急性白血病患者骨髓单个核细胞、巨核细胞白血病CHRF细胞株、人组织细胞淋巴瘤U937细胞株、人慢性粒细胞白血病K562细胞株及10名健康供者外周血单个核细胞内TIMP3基因启动子区域的甲基化情况。结果50例患者骨髓单个核细胞、3种白血病细胞株及健康供者外周血单个核细胞中均未检测到TIMP3启动子区域甲基化。结论TIMP3基因启动子区域甲基化与急性白血病发病可能无相关性。 Objective To investigate the effect of promoter methylation of tissue inhibitor of metalloproteinase-3 (TIMP3) on pathogenesis of acute leukemia(AL). Methods The methylation status of TIMP3 promoter were investigated in bone marrow mononuclear cells of AL patients, CHRF cell lines, U937 cell lines, K562 cell lines and peripheral blood mononuclear cells of healthy volunteers by methylation specific polymerase chain reaction(MS-PCR). Results No methylation was detected in all of bone marrow mononuclear cells of AL patients, three kinds of leukemia cell lines and peripheral blood mononuclear cells of healthy volunteers. Conclusion There may be no relationship between promoter methylation of TIMP3 and the pathogenesis of AL .
