摘要
目的检测鼠疫近缘菌中是否存在着与鼠疫菌pYC质粒相同的核苷酸序列。方法设计4对引物,其产物长度覆盖整个的pYC质粒序列,将产物设计为探针,对近40余种鼠疫近缘菌全部的DNA进行斑点杂交。结果除用来作为对照的、分离至云南的4株鼠疫菌斑点杂交呈阳性外,其他杂交结果均为阴性。结论目前发现该质粒只存在于某些鼠疫菌中,进一步证实了我们先前的猜测,说明该质粒不是从这些近缘菌进化而来,可能是从外界获得的。
Objective To search for whether some bacteria closely related to Yersinia pestis exist the same nucleotide sequence as the plasmid pYC of Yersinia pestis. Methods Four primers were designed with the products covering the whole nucleotide sequence of the plasmid pYC, and the products were used to conduet dot blot hybridization with the whole DNA of these related bacteria. Results The hybrided results were negative except for the four strains of the control from Yunnan Province. Conclusions It is found that the plasmid is only detected in Yersinia pestis. It accords to our previous hypothesis, and indicates that the plasmid is from the related environment but not from the evolution process of Yersinia pestis.
