糖脉平对2型糖尿病大鼠血管纤溶酶原激活抑制剂表达的影响 被引量:1

摘要 目的观察糖脉平对2型糖尿病大鼠血管中纤溶酶原激活抑制剂(plasminogen activeator inhibior,PAI-1)表达的影响,探讨其对血管保护作用的机制。方法Wistar大鼠大剂量链脲佐菌素腹腔注射(STZ60mg/kg),造模成功后随机分为正常对照组、模型对照组、糖脉平组、二甲双胍组、玉泉丸组,每2周称体重并剪尾测血糖,第6周处死取主动脉标本采用免疫组化SABC法测PAI-1的表达。结果造模后,大鼠血糖明显升高,PAI-1的表达明显增高,经治疗后,糖脉平治疗组及二甲双胍组的PAI-1表达明显降低,与糖尿病模型组比较有显著差异(P<0.01)。结论糖脉平能减少糖尿病大鼠PAI-1的表达,减少血管血栓的形成,对糖尿病血管病变起保护作用。 Objective To investigate the effects of Chinese medicine Tang maiping (TMP) on plasminogen activeator inhibitor (PAL-1) in blood vessel endothelium of type 2 diabetic rats. Methods The models of type 2 diabetic Wistar rats were established by injecting high dosage (60 mg/kg) of streptozotocin (STZ), and fifty model rats were randomly divided into normal control group, model control group, Tang maiping group, Metformin group and Yuqanwan group. The body weight and blood sugar by cutting the rats tails were measured ever two weeks. At the sixth week rats were executed and the main arteries were sampled. The PAL-1 expression was assessed by SABC immunohistochemical method. Results For model rats, the blood sugar level rose obviously and the body weight decreased. After post-treatment, PAI-1 expression of Tang maiping group and Metformin group decreased significantly than control group (P〈0. 01). Conclusion Tang maiping recipe can decrease the expression of PAL-1 and the incidence of microthrombus to protect the vascular structure and function in diabetic rats.
