摘要 通过正交设计试验,探讨了超临界CO2法萃取北五味子果实中五味子醇甲的工艺条件,并建立了五味子醇甲的质量分数高效液相色谱测定方法。研究表明:最佳提取条件为萃取压力25MPa,萃取温度45℃,萃取时间60min,提取率可达到0.970%;以乙腈-水溶液为流动相,在Zorbax Eclipse XDB-C18柱进行高效液相色谱分析,平均回收率为98.64%,相对标准偏差为2.908%,检出限为0. The optimal conditions of supercritical carbon dioxide extraction for schizandrin from fruits of Schisandra chinensis (Turcz.) Baill were studied by L9 (3^4) orthogonal test. A high performance liquid chromatography (HPLC) method for determining schizandrin in S. chinensis was established. The optimal extraction conditions were obtained as pressure 25 MPa, temperature 45 degrees C, and extraction time 60 minutes. The extraction ratio of schizandrin reached 0.970%. RP-HPLC analysis was performed on a Zorbax Eclipse XDB-C18 column with a linear solvent methanoL/water gradient. The average recovery was 98.64%, the relative standard deviation was 2.908%, and the detection limit was 0.02μg/L.
