摘要
目的:观察木兰脂素对大鼠肾缺血再灌注损伤时肾组织Toll样受体4(TLR4)表达的影响。方法:采用大鼠肾缺血再灌注损伤模型,雄性SD大鼠45只随机分为假手术组(n=5)、对照组(n=20)和治疗组(n=20)。于再灌注开始前5min静脉注射木兰脂素2mg/kg或等量生理盐水。再灌注达相应时间点时,从各组大鼠取血备测血肌酐(Cr)、血尿素氮(BUN)以及血清细胞间黏附分子-1水平,肾组织用于观察病理变化以及作免疫组化分析肾组织TLR4表达情况。结果:治疗组BUN、Cr较相应时间点对照组明显降低,肾组织损伤也较对照组明显减轻。治疗组肾组织TLR4阳性表达多于假手术组,但较对照组明显减少。结论:木兰脂素预处理可减轻肾缺血再灌注损伤,其机制可能是通过抑制TLR4的表达而发挥作用的。
Objective To observe the effects of magnolin on expression of toll-like receptor 4 (TLR4) in rats with renal isehemia-reperfusion injury. Methods A rat model of renal ischemia-reperfusion injury was established. Rats were randomized to three groups: sham-operation group, ischemia-reperfusion group, and magnolin-treated group. Magnolin at 2 mg/kg or equivalent normal saline was administered intravenously 5 min before reperfusion. Serum levels of ereatine (Cr), blood urea nitrogen (BUN), and intercellular adhesion molecule-1 were detected at different time points, so were the pathologic changes and the expression of TLR4 in the renal tissues. Results As compared with the ischemia-reperfusion group, serum levels of Cr and BUN were markedly declined and the renal injury was relieved in the magnolin-treated group. More TLR4 expressed in the magnolin-treated group than in the sham-operation group, while the TLR4 expression in the magnolin-treated group was significantly decreased, as compared to the ischemia- reperfusion group. Conclusion Magnolin can attenuate renal ischemia-reperfusion injury, whose mechanism may be involved in the inhibition of TLR4 expression.
