摘要
目的:探讨美托洛尔对心力衰竭(心衰)大鼠心肌细胞凋亡的影响。方法:构建心衰大鼠模型,随机分为心衰组、美托洛尔组(各15只)及假手术组(n=15),为同期开胸,无血管结扎,8周后,测定血流动力学指标,观察左右心室质量指数(LVMI.RVMI),TUNEL法检测心肌细胞凋亡率。结果:①与假手术组比较,心衰组血流动力学指标明显恶化,LVMI和RVMI显著升高,心肌细胞凋亡率增加;②与心衰组比较,美托洛尔组,MBP和LVEDP下降,±dp/dtmax均升高,LVMI和RVMI下降,心肌细胞凋亡率下降。结论:美托洛尔能抑制心肌细胞凋亡,改善心功能,逆转心室重构。
Objective: To explore the effect of metoprolol on cardiomyocyte apoptosis in rats with heart failure. Methods: Rat models with heart failure were established and randomly divided into heart failure group (n = 15), metoprolol group (n = 15), and sham-operation group (n = 15). Hemodynamic parameters, left and right ventricular mass index (LVMI, RVMI) were observed, and cardiomyocyte apoptosis were detected by the method of TUNEL in each group after 8 weeks. Results: ① Compared with those of sham operation group, hemodynamie parameters were deteriorated, LVMI and RVMI were elevated significantly and cardiomyocyte apoptosis was increased in heart failure group. ② Compared with those of heart failure group, MBP, LVEDP were descended, ± dp/dtmax elevated, LVMI and RVMI was descended, cardiomyocyte apoptosis decreased in metoprolol group. Conclusions: Metoprolol can inhibit cardiomyocyte apoptosis, improve heart function and reverse ventricular remodeling.
