摘要
采用高效液相色谱法测定人血浆中10α-甲氧基-9,10-二氢麦角醇(MDL,尼麦角林体内代谢物)的浓度.MDL和麦角新碱(内标)用氯仿从血浆中提取,并在SphersorbC18柱上,以乙腈—0.1mol/L(pH5)醋酸铵(14)为流动相进行分离,在225nm处检测.本法简便、灵敏、专属性好.
A method for the determination of 10αmethoxy9,10dihydrolysergol(MDL),a nicergoline metabolite,in human plasma has been developed using HPLC.MDL and ergometrine as internal standard were extracted from the plasma with chloroform and separated on a reversed-phase ODS column with a mobile phase of acetonitrile-01mol/L(pH5)ammonium acetate(1∶4).The eluate was detected at 225nm,the method is simple,sensitive and specific.
