摘要
目的探讨弥漫性脑损伤(DBI)后损伤区脑组织基质金属蛋白酶-9(MMP-9)和γ-氨基丁酸(GABA)的表达。方法采用自由落体撞击法制作成年雄性SD大鼠弥漫性脑损伤模型,大鼠随机分为对照组、伤后3、6、12、24、72h和7d组。采用免疫组化ABC法测定MMP-9和GABA的表达。结果MMP-9在DBI后表达逐渐增强,至伤后24h达到高峰,伤后7d仍保持较高水平;而GABA在DBI后表达逐渐减弱,至伤后12h达到低谷,伤后72h恢复正常。MMP-9和GABA表达时相和强度呈负相关关系。结论DBI可引起损伤区脑组织MMP-9表达增强和GABA的表达减弱,可能在脑组织的继发性损害中起重要作用。
Objective The aim of this study was to investigate the expression of metalloproteinase-9 (MMP-9) and gamma-aminobutypic acid (GABA) in injured brain and to explore potential role of MMP-9 and GABA after diffuse brain injury (DBI). Methods The trauma was produced by a free falling weight on the exposed dura of right parietal lobe. The rats were randomly divided into control group and DBI groups at 3, 6, 12, 24 and 72h, and on 7d. The expression of MMP9 and GABA was examined by ABC immunohistochemistry. Remtlts The results showed that the expression of MMP-9 in the injured site progressively increased, reached the maximum at 24h and kept at high level up to 7d after DBI. While the expression of GABA went down to the lowest level at 12h and up to normal level at 72h after DBI. The levels of MMP-9 and GABA observed in the injured brain showed a high negative correlation. Conclusions It was concluded that cortical contusion trauma could induce a concomitant and persistent upregulation of MMP-9 and downregulation of GABA expression in the injured brain in rats which might play a central role in the secondary injury after DBI.
